Context
This line is written by John in John 3:3.
So this Pharisee guy named Nicodemus comes to talk to Jesus one night, and they have a little conversation about being born again and stuff like that. Nicodemus is a little confused about the whole situation, but Jesus lets him know that the second birth is more of a spiritual thing. Don't worry, Nic, it's not as weird as it sounds.

This is one of the more famous quotes in the Bible, and being "born again" is one of the most important parts of a lot of Christian religions (it's where the term "born again Christian" comes from).
